Design Notes
We remove all forbiden restriction sites, (EcoRI,XbaI,SpeI,PstI), from P4 genome but it means remove the integrase also. For our project it was useful because removing the integrase gene we expect P4 to be maintained as a plasmid. However it could mean a reduction in the stability of the plasmid without selection.
